RESEARCH PROJECTS on “Agri Biotechnologies”
for B.Sc. / B.Tech / M.Sc. students
 
Applications are invited for theme based project/dissertation works on Plant molecular biology (Genetic engineering and Molecular Marker assisted breeding), plant tissue culture and Microbial biotechnology
 
Agri Biotech Foundation is an autonomous Institution engaged in research, extension and training. It is located on Prof. Jayashankar Telangana State Agriculture University Campus, Rajendranagar, Hyderabad. ABF has been recognized as Scientific and Industrial Research Organization (SIRO) by the Department of Scientific and Industrial Research (DSIR), New Delhi.
 
Themes of dissertations: Dissertations can be taken up on the following themes
 
Theme 1. Genetic engineering:
 
Gene Cloning: Isolation of gene, promoter, Restriction analysis, ligation, cloning in different vectors (TOPO-TA, plant expression vectors), DH5α transformation, PCR analysis
 
Agrobacterium mediated plant transformation: Mobilization of plant expression vectors into Agrobacterium by freeze thaw method and electroporation, explant preparation, co-cultivation, differentiation of transformed and non transformed explants, optimization of regeneration of shoots and roots in different hormonal growth media, orientation on plant growth conditions at glass house.   
 
Characterization of biotechnologically developed plants
PCR analysis for identification of transformed and non transformed plants, Southern blot analysis for detection of copy number of transgenes, training on different stress impositions on plants, sample collection, Biochemical and phenotypic characterization, protein detection through strip assays.
 
Gene expression studies: RNA isolation, cDNA synthesis, quantitative transcript analysis of gene
 
Theme 2. Marker assisted breeding
Tagging and mapping gene(s) of interest in rice/ blackgram
Development of mapping populations
Phenotyping for resistance to bacterial blight, blast, BPH, WBPH or gall midge
Genotyping with SSR, EST-SSR markers
Linkage analysis and QTL mapping
Identification and functional validation of candidate gene
 
Theme 3. Plant Tissue culture
Standardization of regeneration protocols for various species through organogenesis and somatic embryogenesis.
Techniques to be covered: Nutrient media preparation, various sterilization methods, Explants isolation and inoculation, multiplication through sub culturing, in vitro & in vivo rooting and acclimatization.
 
Theme 4. Microbial biotechnology
Genomic DNA from Gram+ve, Gram–ve bacteria and fungi;
Bar coding in microbial genomics;
Cloning and transformations techniques in Plant-micro interactions;
Fluorescence image sensing techniques;
Transcriptomic studies in plant-microbes;
Bacterial protein profile, quantitative and qualitative analysis-SDS and Western blotting; Plant probiotics,
Piolet scale fermentation techniques
Chromatographic techniques (Column, TLC, HPLC)
 
Course fee: It is based on duration of the project
 
Eligibility: B.Sc., B.Tech. M.Sc., any life sciences
